G-CSF and SCF in combination can stimulate granulocytic differentiation and proliferation of CD34+ human haemaopoietic stem cells. G-CSF is glycosylated with one O-linked oligosaccharide. SCF is heavily glycosylated with up to 50% of its molecular weight consisting of carbohydrate.
Apollo's G-CSFhcx and SCFhcx were evaluated in a colony-forming assay, whereby CD34+ cells were inoculated into semisolid cultures of methylcellulose with G-CSF and SCF and grown for 14 days. Apollo's glycosylated G-CSFhcx and SCFhcx produced more and larger colonies, compared to commercially available non-glycosylated cytokines produced in E. coli as illustrated below.
When used in combination to expand haemtopoietic stem cells, Apollo's glycosylated G-CSFhcx and SCFhcx also promoted significantly more cell proliferation (up to 53% higher: p<0.0001) than E. coli expressed G-CSF and SCF.
This study shows that hcxTM G-CSF and SCF used together form a powerful combination for inducing stem cell expansion in vitro. As proteins are expressed in human cell lines this gives them unique potential for clinical therapies, where there is a growing preference for eliminating non-human products.
